A Tory MP told Theresa May to her face that party activists want her to quit within the next week.
Sir Peter Bone said members in his Wellingborough and Rushden constituency want the Prime Minister to step down before the European Parliament elections on 23 June.
They argued that the deal she struck with the EU was worse than staying in, and demanded the country leaves the bloc without any agreement at all.
Mrs May thanked the activists for their help and insisted the Government was trying to deliver Brexit but that MPs were thwarting the process.
Mr Bone delivered the message during Prime Minister’s Questions - the most high-profile session in the parliamentary timetable - in a humiliating blow to the Tory leader.
He said he had joined some members on Saturday as they went door-knocking ahead of polling day, and that they had given him a letter to pass to the Prime Minister.
“They say that her deal is worse than staying in the European Union; that they want us to come out now on a no-deal basis,” he explained.
“More importantly, they've lost confidence in the Prime Minister and wish her to resign before the European elections. Prime Minister: what message do you have to say to these dedicated and loyal Conservatives?”
Mrs May shot back: “This is a government that wants to deliver Brexit and has been working to deliver Brexit.
“Sadly, so far, the House of Commons has not found a majority to do that.
“If everybody in the House of Commons had voted alongside with the Government and the majority of Conservative members of Parliament, we would already have left the European Union.”
It comes after Mrs May announced that her deal would be put to the Commons once more in the first week of June in the form of the Withdrawal Agreement Bill.
It has already been rejected three times, most recently by a majority of almost 60. Tory MPs especially are against the so-called 'backstop' plan to protect the Northern Irish border.
The Prime Minister will meet with senior party bosses this afternoon who are expected to demand she lays out a timetable for here departure.
